Basic Helix Loop Helix (bHLH) is a common eukaryotic regulatory protein structural motif. bHLH protein plays important role in the gene expression during the developmental process, however exact mechanism of their interplay with other transcription factors and adaptability of the structure is not well understood.
E47-NeuroD1/Beta2 bHLH Domain-DNA Complex displays dimerization and DNA binding specification. It’s composed of 2 classes each with 2 alpha helices separated by a loop. E47 is the class I factor, known to regulate B lyphocytes development. Neuro D1 is the class II factor, known to regulate endocrine related genes as well as neuronal differentiation during development and hormon precursor. Mutation in human neuroD1 gene can lead ataxia, deafness and diabetes.
Understanding the structure of protein DNA binding complex allows us to further study mutations of bHLH proteins and understand its minimal binding configuration.
