ZFP568 is a tandem Krüppel associated box (KRAB) zinc finger protein that functions as a repressor of placental specific insulin like growth factor 2 (IgF2-P0) expression in mice. It contains three main component sections; 2 N terminal KRAB domains for transcriptional repression and an 11 zinc finger (ZnF) array at the C terminal. ZFP568 ZnFs bind to the 24 nucleotide IgF2-P0 DNA consensus sequence, where the recruitment of corepressor KAP1 establishes silencing through H3K9 methylation. ZFP568 recognises the 24-nucleotide motif through a shortened DNA minor groove created by an AT rich sequence. Each ZnF forms different interactions with the IgF2-P0 consensus sequence, contacting 2, 3 or 4 nucleotide bases on the same or opposite strand. ZFP568:DNA is not well conserved organisms such as humans due to evolutionary mutations.
